Go to Launcher -> Options -> PR Mumble tab and make sure your key binds are set correctly. It's also good idea to remove key binds from mute and volume controls.

Then launch Mumble from the same tab. In Mumble, go to Configure -> Audio Wizard... and test different input and output devices until you can hear male voice speaking, and input volume meter reacts to your voice. After finishing the wizard, quit Mumble from Server -> Quit Mumble menu or from Mumble icon next your task bar clock.
